Studies on some physiological behaviour of local Azospirillum inoculantes

The present study was conducted with a few isolate and characterizes rhizostheric Azospirillum form roots of grass and non-legume gains crops prevalent in Ranchi and Gumla districts. The isolates were also evaluated for their ability to fix nitrogen and to synthesize planned regulator substance. Five isolates of Azospirillum were isolated from roots of maize, sugarcane, gora dhan, kodo and rice. Morphological features of the isolates of Azospirillum, revealed as short vibrioid gram negative cells. Isolates ofAzospirillum developed as sub-surface pellicle (1-2 mm below the surface) in the nitrogen free semi solid malate medium. However when grown under aerated condition in presence of combine nitrogen, they exhibited thin, converse and rugose colonies on Okon’s agar plates and showed moderate growth effuse form on agar stroke. The isolates belonging to the genus invariable exhibited a positive response to catalase. Growth of Azospirillum as sub-surface pellicle in nitrogen free semi-solid malate medium indicated their low requirement of oxygen under nitrogen fixing condition. Among Azospirillum isolates ASS -2 and ASG-3 were more efficient in nitrogen fixation (13.40 mg and 13.00 mg) N ml-1 in culture medium respectively) under laboratory conditions. The use of different carrier materials like vermin compost+soil and FYM+soil revealed equal viability up to one month. Huge possibility may be predicted in the content of biofertilizers after seeing market survey and rural extension programme.
